چکیده

PREVIOUS studies on the effect of digitalis on the hypothermic heart have indicated that the arrhythmic effects of hypothermia and digitalis are antagonistic. Those experiments were primarily intended to study the effect of digitalization on hypothermia using pharmacological doses of digitalis. In the experiments reported herein, the procedure was reversed in that it was thought to be of value to investigate the possible use of hypothermia as an antagonist of digitalis toxicity. For this purpose dogs injected with toxic doses of ouabain were subsequently cooled and stabilized in a state of moderate hypothermia at body temperatures of 28 to 31 C. where spontaneous fibrillation (due to hypothermia) does not occur. The clinical problem of digitalis toxicity is too well known and needs no further comment. However, it should be emphasized at this point that digitalis-induced arrhythmias are only a prelude and warning of impending death due to ventricular fibrillation. Yet arrhythmias and ventricular fibrillation are not entirely dependent on the same mechanisms, so that protection from arrhythmias is not necessarily associated with protection from death due to ventricular fibrillation or possibly cardiac standstill. It is essential, therefore, to determine whether hypothermia could alter the lethal as well as the arrhythmic effects of digitalis.

Effect of Hypothermia on the Tolerance of Dogs to Digitalis

H modifies to a considerable degree the effects of various drugs. Among those investigated in the past have been central nervous system stimulants and depressants, hormones, epinephrine, acetylcholine, insulin, thyroid, ethyl alcohol, Dicumarol, oxygen, carbon dioxide, procaine, and cocaine. However, the effect of hypothermia on the action of digitalis has not been extensively investigated. Mos...

Digitalis cardiotoxicity.

Clinical manifestations of digitalis toxicity were clearly described by Withering in 1785. One hundred years later, certain digitalis-induced arrhythmias were inscribed on the smoked drum, and shortly thereafter with the introduction of the electrocardiograph, manifestations of digitalis toxicity as recognized today were recorded in animals and human beings. With popularization of the direct-wr...

EFFECTS OF CHOLECYSTOKININ RECEPTOR ANTAGONISTS ON MORPHINE- AND COCAINEINDUCED HYPOTHERMIA

The effects of cholecystokinin (CCK) receptor antagonists on hypothermia induced by cocaine or morphine have been studied in mice. In the present work, subcutaneous (SC) injection of cocaine (50-150 mg/kg) or morphine (125-500 mg/kg) induced hypothermia in mice. Administration of CCKA receptor antagonist MK-329 (0.5-1.5 mg/kg), CCKB receptor antagonist L-365, 260 (0.5-1.5 mgl kg) and CCK r...

Cardioversion and digitalis. IV. Effect of beta adrenergic blockade.

Experiments were performed in anesthetized dogs to study some effects of propranolol, dextro-propranolol and ICI50,172 on digitalis-induced arrhythmias and digitalis-induced sensitization to electrical shock. When given in beta-blocking doses, propranolol did not abbreviate ouabain-induced ventricular tachycardia (VT) or decrease ouabain-induced sensitization to electrical shock. In larger dose...

Background and Aim: Many studies have shown the neuroprotective effect of systemic hypothermia in the treatment of spinal cord injury. But the effect of delay hypothermia is not known. The goal of this study was to evaluate the effects of Methylprednisolone on the therapeutic window of hypothermia treatment following experimental Spinal Cord Injury (SCI) by measuring the accumulation of Poly...
